Abstract

Objective To explore the therapeutic mechanism of Zizhong Yishen Pill(子仲益肾丸,ZZYSP)in the treatment of oligoas‐thenospermia(OAS)by observing its effects on the expression of related proteins and mRNA in rat testicular tissue.Methods Thirty male SD rats were randomly divided into a blank control group(n=10)and a pre-modeling group(n=20).The pre-modeling group was treated with dibutyl phthalate(DBP)to establish an OAS rat model,after which they were randomly assigned to a model group and a ZZYSP group.The rats were administered deionized water or a ZZYSP suspension via gavage for four weeks.Semen quality and repro‐ductive hormone levels were evaluated in each group.Pathological changes in testicular tissue were observed using HE staining,while Western blot(WB)and q-PCR were employed to detect the expression of PI3K/AKT,BAD,and Bcl-2 proteins and their corresponding mRNAs in testicular tissue.Results Compared with the blank control group,the model group exhibited significantly decreased sperm concentration and motility,along with an increased sperm deformity rate(P<0.05),whereas the ZZYSP group showed marked improve‐ment in sperm quality(P<0.05).Serum testosterone(T)levels were significantly reduced in the model group(P<0.0001),while follicle-stimulating hormone(FSH)and luteinizing hormone(LH)levels were significantly elevated(P<0.0001,P<0.001,respec‐tively).In contrast,the ZZYSP group demonstrated significantly increased serum T levels(P<0.01)and decreased FSH and LH levels(P<0.001,P<0.05,respectively).HE staining revealed testicular tissue damage and increased apoptotic cells in the model group,whereas pathological injury was alleviated,and apoptotic cells were reduced in the ZZYSP group.WB and q-PCR analyses indicated that the model group exhibited significantly downregulated expression of PI3K,AKT,and Bcl-2 proteins and mRNAs,along with upregulated BAD protein and mRNA expression.Conversely,the ZZYSP group showed significantly upregulated PI3K,AKT,and Bcl-2 expression and downregulated BAD expression.Conclusion ZZYSP may treat OAS by inhibiting spermatogenic cell apoptosis in testes through modulation of PI3K/AKT signaling pathway.
