Molecular Characterization of Two theta Class Glutathione-S-Transferase Genes and Their Response to Environmental Stress in Myzus persicae

Abstract
To elucidate the molecular characteristics of theta class glutathione S-transferases(GSTs)in Myzus persicae and reveal their function in response to environmental stress,the cDNA sequences of two theta class GST genes(designated MpGSTt1 and MpGSTt2)were identified from the tran-scriptome of M.persicae.The sequence features of MpGSTt1 and MpGSTt2 were analyzed using bioinformatic softwares.Real-time quantitative PCR was used to investigate the transcriptional pro-files of two genes during different development stages of M.persicae and the changes of their mRNA levels under various environmental stress.The results showed that the open reading frames of MpG-STt1 and MpGSTt2 genes were both 699 bp,and the proteins encoded by the two genes both contained 232 amino acid residues with a molecular mass of 27.3 ku.The transcriptional levels of MpGSTt1 and MpGSTt2 can be detected at different developmental stages of the tested M.persicae,and the highest levels were found at the 3rd instar nymphs.The transcription levels of MpGSTt1 and MpGSTt2 reached the highest level at 0.5 h after exposure to high temperature(35℃),which were 1.63-and 3.72-fold,respectively,higher than the levels of MpGSTt1 and MpGSTt2 in controls.Similarly,the transcription levels of MpGSTt1 and MpGSTt2 were significantly at 0.5 and 1 h,respectively,upregu-lated after exposure to ultraviolet-B(UV-B),which were 3.46-and 1.95-fold,respectively,higher than the levels of MpGSTt1 and MpGSTt2 in controls.The transcription levels of MpGSTt1 and MpGSTt2 in M.persicae were significantly increased after treatment with lambda-cyhalothrin and Lecanicillium psalliotae.MpGSTt1 and MpGSTt2 reached their highest mRNA levels at 18 h and 6 h,respectively,after lambda-cyhalothrin treatment,whereas MpGSTt1 and MpGSTt2 reached their highest transcription levels at 48 h and 24 h,respectively,after L.psalliotae treatment.The results of this study indicate that MpGSTt1 and MpGSTt2 may play essential roles in the adaptation of M.per-sicae to environment.关键词
