摘要

Abstract

Objective It is to investigate the interventional effects and related molecular mechanisms of decoction for reinforcing healthy Qi and removing toxin(DRHQRT)on cisplatin in the treatment of Lewis lung cancer tumor-bearing mice.Methods Thirty-five C57BL/6J male mice were selected,in which 7 mice were randomly selected as blank group(not modeled),and the remaining 28 mice were subcutaneously inoculated with Lewis cells to replicate models of Lewis lung carcinoma.The successfully modeled mice were randomly divided into model group,cisplatin group,DRHQRT group,and cisplatin+DRHQRT group,with 7 mice in each group.The cisplatin group was given cisplatin 2 mg/kg via intraperito-neal injection(twice weekly),the DRHQRT group was given DRHQRT decoction solution 0.2 mL by gavage(once dai-ly),the cisplatin+DRHQRT group were given cisplatin 2 mg/kg via intraperitoneal injection(twice weekly)and DRHQRT decoction solution 0.2 mL by gavage(once daily),the model group and control group were given an equal vol-ume of physiological saline by gavage,all groups were administered continuously for 14 days.The tumor weight of trans-planted tumors was recorded,tumor suppression rate and splenic index were calculated,the proportions of CD3+,CD4+and CD8+T-cells in spleen tissue were detected by flow cytometry,and serum levels of interleukin-6(IL-6),transforming growth factor-β(TGF-β),vascular endothelial growth factor(VEGF)were measured by ELISA,the expressions of pro-teins related to nuclear factor-κB(NF-κB)signaling pathway in tumor tissues were determined by Western blot.Results The tumor weights of mice in all treatment groups were significantly lower than those in the model group(all P<0.05),and the tumor weights of mice in cisplatin+DRHQRT group was significantly lower than that in the cisplatin group(P<0.05);the tumor suppression rate in the cisplatin+DRHQRT group was significantly higher than that in the cisplatin group(P<0.05).Compared with the blank group,the proportions of CD3+,CD4+and CD8+T cells were significantly de-creased,while the serum levels of IL-6,TGF-β and VEGF were significantly increased in the model group(all P<0.05).Compared with the model group,the proportions of CD4+and CD8+T cell in the cisplatin group and the proportions of CD3+,CD4+and CD8+T cell s in the cisplatin+DRHQRT group were significantly increased(all P<0.05),the serum levels of IL-6,TGF-β,and VEGF were significantly reduced in cisplatin group and cisplatin+DRHQRT group(all P<0.05);the proportions of CD4+and CD8+T cells in the cisplatin+DRHQRT group were significantly higher than those in the cisplatin group(all P<0.05),while the proportions of CD3+,CD4+and CD8+T cells were significantly higher than those in the DRHQRT group(P<0.05);the serum levels of IL-6,TGF-β and VEGF in the cisplatin+DRHQRT group were significantly lower than those in the cisplatin group(all P<0.05),and serum level of IL-6 was significantly lower than that in the DRHQRT group(P<0.05).The protein relative expressions of A20 and cyclin D1 in transplanted tumor tissues were significantly lower in all treatment groups than those in the model group(all P<0.05);the protein relative expressions of p65 in transplanted tumor tissues were significantly lower in DRHQRT group,and cisplatin+DRHQRT group than that in the model group(all P<0.05).Conclusion Decoction for reinforcing healthy Qi and removing toxin combined with cisplatin is more effective than cisplatin alone in inhibiting growth of transplanted tumors,and can alleviate cisplatin-induced damage of immune organs,and regulate balance of immune cells in Lewis lung cancer tumor-bearing mice,and its antitumor mechanism may be related to regulation of NF-κB signaling pathway.
