摘要

Abstract

Objective It is to investigate the effects of modified Bupleurum Powder for Soothing Liver(MBPSL)on coronary artery spasm based on the RhoA/Rho Kinase signaling pathway.Methods Seventy-two adult SD rats of SPF grade were selected,in which 12 rats were randomly divided into normal group,while the remaining 60 rats were injected with posterior pituitary hormone via tail vein to establish models of coronary artery spasm.The successfully modeled rats were randomly divided into model group,fasudil group,high-dose,medium-dose,and low-dose groups of MBPSL,with 12 rats in each group.The fasudil group was given Fasudil 15 mg/kg by intraperitoneal injection,the high-dose,medium-dose,and low-dose groups of MBPSL were respectively given modified MBPSL 7.04 g/kg,3.52 g/kg,and 1.76 g/kg by gavage,the normal group and model group were given an equal volume of distilled water by gavage,all administered once daily,continuously treated for 4 weeks.The number of leads showing ST-segment depression,T-wave flattening,and T-wave in-version in electrocardiograms(ECGs)at the end of modeling and treatment were recorded in all groups(except for the nor-mal group),the ECG efficacy of each group after treatment was assessed,the mRNA expressions of Rho kinase,RhoA,myosin phosphatase target subunit 1(MYPT1)in myocardial tissue were detected by Real-Time PCR,the protein expres-sions of RhoA,RhoA-GTP,MYPT1 and p-MYPT1 in myocardial tissue were detected by Western blot.Results After treat-ment,the number of leads showing ST-segment depression,T-wave flattening,and T-wave inversion in the fasudil group,high-dose MBPSL group,and medium-dose MBPSL group were all significantly fewer than those in the model group(all P<0.05),the ECG total effective rate was significantly higher than that of the model group(P<0.05).The mRNA relative expressions of Rho kinase,RhoA,MYPT1,as well as the protein relative expressions of RhoA,RhoA-GTP,MYPT1,and p-MYPT1 in myocardial tissue in the model group were significantly higher than those in the normal group(all P<0.05),the mRNA relative expressions of Rho kinase,RhoA,MYPT1,as well as the protein relative expressions of RhoA,RhoA-GTP,MYPT1,and p-MYPT1 in myocardial tissue in the fasudil group,high-dose MBPSL group,and medium-dose MB-PSL group were significantly lower than those in the model group(all P<0.05),but there were no statistically significant differences in these indicators among the first three groups(all P>0.05).Conclusion Modified Bupleurum Powder for Soothing Liver can reduce coronary artery spasm via inhibiting the RhoA/Rho kinase signaling pathway.
