基于Super-GBS测序的非洲菊种质遗传多样性与遗传结构分析
Genetic structure and diversity analysis of Gerbera jamesonii germplasm based on Super-GBS sequencing

Abstract
[Objective]The paper aimed to analyze the genetic structure and diversity of 151 gerbera(Gerbera jamesonii)germplasms repre-senting seven distinct types,providing a foundation for the introduction,conservation,and breeding utilization of gerbera germplasm resources.[Method]A total of 151 gerbera germplasms were subjected to genome-wide SNP detection using Super-GBS sequencing technology.Genetic diversity parameters,genetic distances,principal components analysis(PC A),phylogenetic evolution and genetic structure were analyzed based on the obtained high-quality SNPs.[Result]A total of 604 216 282 clean reads with 88.2 Gb valid data were obtain.The constructed reference genome contained 269 599 sequences,yielding 41 686 high-quality SNPs after filtration.Across gerbera types,observed heterozygos-ity(Ho)ranged from 0.1419 to 0.1888,expected heterozygosity(He)from 0.1401 to 0.2293,polymorphic information content(PIC)from 0.1115 to 0.1888,nucleotide diversity(Pi)from 0.1652 to 0.2289,observed allele number(Na)from 1.3675 to 1.971,and effective al-lele number(Ne)from 1.2420 to 1.3630.Pasta,bubble and potting gerberas exhibited lower genetic diversity,whereas standard and com-pound colored gerberas showed higher diversity.Genetic distances between groups ranged from 0.0047 to 0.2738,with the smallest distance(closest relationship)between standard and compound colored types,and the largest(most distant)between pasta and potting types.Phylo-genetic and genetic structure analyses classified the germplasms into five clusters:Potting gerberas formed a distinct group;Pasta,bubble and other single-source germplasms clustered closely,while standard,compound colored and spider types displayed dispersed distributions.[Con-clusion]Significant genetic diversity differences exist among gerbera germplasm types.Germplasms of identical types with singular origins ex-hibit lower diversity and closer kinship,while diverse types segregate into five clusters.These findings support strategic germplasm introduc-tion,conservation and enhanced breeding efficiency in gerbera.关键词
