摘要

Abstract

Objective To identify the targets of taraxerol(TAX)in the treatment of colorectal cancer(CRC),so as to provide theoretical basis for new treatment strategies.Methods PharmMapper and SwissTarget Prediction were used to pre-dict the therapeutic targets of dandelion terpenoids,and standardization was performed by UniProt.The GSE83129 dataset was downloaded from the GEO database,and limma package was used to screen the differentially expressed genes(DEGs).The therapeutic target of TAX was intersected with the DEGs by Venny,and the target of TAX for the treatment of CRC was obtained.And the protein-protein interaction network(PPI network)was constructed through STRING and Cytoscape to screen the hub genes.The KEGG signaling pathways of hub genes were identified through GO/KEGG enrichment analysis,and the structure of the hub genes was obtained from PDB,and the molecular docking test with TAX was conducted by AutoDock Tools.Finally,the docking results were visualized through PyMOL.Results Through the PPI network,32 hub genes were identified,including HSP90AB1,HSP90AA1,and CDK2,etc.Signaling pathways such as PI3K-Akt,p53,nod-like receptors(NLRS),and interleukin-17(IL-17)might be involved in the pathogenesis of colorectal cancer(CRC).Conclusion TAX can target the HSP90AB1,HSP90AA1,CDK2 genes,as well as the PI3K-Akt signaling pathway,p53 signaling pathway,NOD-like receptor signaling pathway and IL-17 signaling pathway to treat colorectal cancer,which indi-cates that TAX can be used as a potential drugs for the treatment of CRC,laying a theoretical foundation for the subsequent verification of therapeutic effects.
