枸杞多糖缓解铝致Sertoli细胞毒性的机制研究
Study on the mechanism of lycium barbarum polysaccharides in the relief of aluminum induced Sertoli cell toxicity

Abstract
Objective To investigate the toxic effects of aluminum exposure on Sertoli cells(TM4 cell line)and its under-lying mechanisms,and to evaluate the alleviating effect of lycium barbarum polysaccharides(LBP).Methods TM4 cells were divided into four groups:normal control(NC)group,aluminum exposure(Al)group,LBP control(NC+LBP)group,and LBP intervention(Al+LBP)group,and an aluminum toxicity model was established.Cell viability was detected by CCK-8 assay,apoptosis rate by flow cytometry,reactive oxygen species(ROS)levels by fluorescence microscopy,and the protein expressions of glial cell line-derived neurotrophic factor(GDNF),Catalase,superoxide dismutase 2(SOD2),Bcl-2,BAX,Cleaved Caspase-9,and Cleaved Caspase-3 by Western blot.Results Compared with the NC group,the Al group showed a decrease in cell viability,an increase in apoptosis rate,an increase in ROS levels and the protein expressions of BAX,Cleaved Caspase-9,and Cleaved Caspase-3,and a decrease in the protein expression of GDNF,Catalase,SOD2,and Bcl-2(all P<0.05).Compared with the Al group,the Al+LBP group showed an increase in cell viability,a decrease in apoptosis rate,a decrease in ROS levels and pro-apoptotic protein expression,and an increase in GDNF and antioxidant/anti apoptotic protein expression(all P<0.05).Conclusion Aluminum exposure damages Sertoli cells by inducing oxidative stress,downregulating GDNF expression,inhibiting cell viability,and promoting apoptosis.Conversely,LBP can alleviate aluminum induced Sertoli cell toxicity by exerting antioxidant and anti-apoptotic effects,while maintaining GDNF expression and cellular viability.关键词
