摘要

Abstract

Objective To investigate the toxic effects of aluminum exposure on Sertoli cells(TM4 cell line)and its under-lying mechanisms,and to evaluate the alleviating effect of lycium barbarum polysaccharides(LBP).Methods TM4 cells were divided into four groups:normal control(NC)group,aluminum exposure(Al)group,LBP control(NC+LBP)group,and LBP intervention(Al+LBP)group,and an aluminum toxicity model was established.Cell viability was detected by CCK-8 assay,apoptosis rate by flow cytometry,reactive oxygen species(ROS)levels by fluorescence microscopy,and the protein expressions of glial cell line-derived neurotrophic factor(GDNF),Catalase,superoxide dismutase 2(SOD2),Bcl-2,BAX,Cleaved Caspase-9,and Cleaved Caspase-3 by Western blot.Results Compared with the NC group,the Al group showed a decrease in cell viability,an increase in apoptosis rate,an increase in ROS levels and the protein expressions of BAX,Cleaved Caspase-9,and Cleaved Caspase-3,and a decrease in the protein expression of GDNF,Catalase,SOD2,and Bcl-2(all P<0.05).Compared with the Al group,the Al+LBP group showed an increase in cell viability,a decrease in apoptosis rate,a decrease in ROS levels and pro-apoptotic protein expression,and an increase in GDNF and antioxidant/anti apoptotic protein expression(all P<0.05).Conclusion Aluminum exposure damages Sertoli cells by inducing oxidative stress,downregulating GDNF expression,inhibiting cell viability,and promoting apoptosis.Conversely,LBP can alleviate aluminum induced Sertoli cell toxicity by exerting antioxidant and anti-apoptotic effects,while maintaining GDNF expression and cellular viability.
