UXS1在肝细胞癌中的表达及临床意义
Expression and clinical significance of UXS1 in hepatocellular carcinoma

Abstract
Objective To investigate the abnormal expression and clinical significance of UDP-glucuronate decarboxylase 1(UXS1)in hepatocellular carcinoma(HCC).Methods Bioinformatics analysis was employed to preliminarily explore the potential functional mechanisms of UXS1 in HCC and its prognostic value,and expression differences between HCC tissues and adjacent tissues were verified through real-time fluorescence quantitative PCR(qPCR)experiments.Results Analysis of the TCGA-LIHC database found that UXS1 was significantly upregulated in HCC.Multivariate Cox regression analysis showed that UXS1 was an independent risk factor,while GSVA analysis suggested that UXS1 might participate in tumor progression by regulating the PI3K-Akt-mTOR signaling pathway,and significantly enriched small molecule metabolism,amino acid metabolism,and signal release in biological processes.The ROC curve confirmed the diagnostic efficacy of UXS1 in distinguishing cancer from adjacent cancer(AUC=0.96),and the column chart constructed based on UXS1 and clinical features of a multifactorial Cox model predicted the survival risk at 1,2,and 3 years(C-index=0.66),which was validated through calibration and decision curves.QPCR experiments showed that UXS1 was significantly upregulated in HCC(P=0.011).Conclusion This study is the first to reveal that UXS1 may promote HCC progression through a metabolism-signa-ling pathway synergy mechanism.It provides a theoretical basis for UXS1 as a prognostic biomarker and therapeutic target,and offers new directions for exploring novel therapeutic strategies for HCC.关键词
