Research progress on the effects of microgravity on digestive system and pharmacokinetics

Abstract
Microgravity is a core physical factor in the special space environment.By causing the redistribution of body fluids,changes in mechanical loading,and alterations in neuroendocrine regulation,it can lead to various physiological imbalances in astronauts,such as gastrointestinal dysfunction and abnormal drug metabolism.During spaceflight,astronauts often experience digestive system disorders,including delayed gastric emptying and impaired intestinal barrier function,along with pharmacokinetic changes such as accelerated drug absorption and reduced metabolic efficiency.These changes pose significant challenges to precise medication administration and nutritional intervention strategies during on-orbit missions.This paper systematically reviews the key mechanisms by which microgravity affects digestive system function and pharmacokinetic processes(including absorption,distribution,metabolism,and excretion)and evaluates the applicability and limitations of relevant ground-based simulation methods.The results indicate that weightlessness can exacerbate cascading disruptions between the digestive system and pharmacokinetics through pathways such as downregulating the expression of intestinal tight junction proteins,disrupting intestinal microbiota homeostasis,and inhibiting hepatic metabolic enzyme activity.A deeper understanding of these mechanisms is of great significance for enhancing the on-orbit health support of astronauts.关键词
