麦角硫因对药物性肝损伤及肝纤维化小鼠的保护作用研究
Protective effects of ergothioneine on drug-induced liver injury and liver fibrosis in mice

Abstract
Objective To investigate the protective effects of ergothioneine on drug-induced liver injury and liver fibrosis in mouse models,and to provide experimental evidence for its development as a potential therapeutic agent.Methods SPF-grade C57 mice were randomly divided into groups.The normal control group received no treatment,while the other groups were odeled according to the experimental design and administered drug interventions based on the rouping plan.The drug-induced liver injury model was established by intragastric administration f acetaminophen solution(300 mg/kg)for three consecutive days until the experimental ndpoint.The liver fibrosis model was established by intraperitoneal injection of carbon etrachloride solution(2 mL/kg).After model establishment,the mice were anesthetized and amples were collected to measure blood and liver-related biochemical indicators,and paraffin ections were prepared to observe pathological changes in the liver tissue of each group.Results n the drug-induced liver injury model,after model establishment,serum ALT and AST levels of mice ncreased significantly,while the liver index decreased markedly,indicating obvious liver njury.Forty-eight hours after model establishment,compared with the model control roup,ergothioneine administration reduced ALT and AST levels,with results consistent with those f the positive drug group treated with diammonium glycyrrhizinate and glutathione.In the liver ibrosis model,obvious fibrous hyperplasia was observed in the liver tissue of mice after model stablishment.Compared with the model control group,the ergothioneine intervention group and he positive drug group treated with diammonium glycyrrhizinate and glutathione showed ignificantly reduced levels of ALT,AST,IL-6,and ALP.HE staining results revealed mild fibrous yperplasia,bile duct hyperplasia,and pigment deposition in the model control roup.However,there were no significant differences in the severity of liver lesions between the rgothioneine-treated groups and the model control group.Conclusion Ergothioneine may mprove ALT and AST levels through its antioxidant mechanism and reduce the expression levels f IL-6 and ALP via its anti-inflammatory effects,thereby demonstrating protective effects in both acetaminophen-induced liver injury and carbon tetrachloride-induced liver fibrosis models.关键词
