基于生物信息学分析的原发性肺腺癌中枢基因鉴定及预后价值分析
Identification and Prognostic Value of Hub Genes in Primary Lung Adenocarcinoma Based on Bioinformatics Analysis

Abstract
Objective To identify key genes as diagnostic and prognostic indicators for lung adenocarcinoma(LUAD).Methods The data set GSE136043 obtained from the GEO database identified differentially expressed genes(DEGs)between LUAD tissues and healthy lung tissues.MetScape was used for Kyoto Encyclopedia of Genes and Genomes(KEGG)and Gene Ontology(GO)enrichment analysis,and SRplot was used to visualize the results.Protein-protein interaction(PPI)module analysis was performed using the Cytoscape MODE plug-in,and 10 hub genes were identified using the Cytohubba plug-in.The expression level of hub genes was verified using the University of Alabama at Birmingham Cancer Data Analysis Portal(UALCAN).Clinical samples were also used to verify three of the key genes.The Gene Expression Profile Interaction Analysis(GEPIA)data set was used to further explore the expression levels of key genes in different pathological stages.Kaplan-Meier plotter was used to analyze the survival of central genes.Results A total of 1049 DEGs(520 up-regulated and 529 down-regulated)were identified.Ten key genes(TOP2A,RRM2,TTK,CDCA8,CDCA3,KIF2C,CDC45,CDC25C,RAD54 Land DTL)were identified from the PPI network.UALCAN validation showed that the expression levels of 10 hub genes were up-regulated.In addition,the overexpression of three hub genes:TOP2A,KIF2C and CDC45 was verified by clinical samples.The increased expression of each hub gene was associated with a lower overall survival rate in LUAD patients.Conclusion TOP2A,RRM2,TTK,CDCA8,CDCA3,KIF2C,CDC45,CDC25C,RAD54L and DTL may be promising prognostic biomarkers and therapeutic targets for LUAD.关键词
