摘要

Abstract

Objective To investigate the effect of fibroblast growth factor 18(FGF18)on the proliferation and differentiation of pre-osteoblasts and the role of forkhead box protein O1(FOXO1)in this process in vitro.Methods CCK-8 assay was used to detect the effect of FGF18 on proliferation of MC3T3-E1 cells.MC3T3-E1 cells were cultured under mineralizing conditions with or without recombinant human FGF18 for 7 days and 21 days.Alkaline phos-phatase staining and alizarin red staining were performed to evaluate the differentiation and mineralization,respectively.Total RNA was isolated and q-PCR was performed to detect the expression of differentiation-related genes.After interfering with Fgf18-shRNA adenovirus,alkaline phosphatase staining was executed following the induction for seven days.Western blot was used to detect FOXO1 and its phosphorylation at different stages of differentiation.Results FGF18 promoted the proliferation while inhibited the differentiation and mineralization of pre-osteoblasts both in a dose-dependent way.After in-terfering with the Fgf18-shRNA adenovirus,osteoblast differentiation was significantly improved than the blank shRNA ad-enovirus control group.Protein electrophoresis analysis revealed that total FOXO1 had no considerable change,while p-FOXO1 was markedly up-regulated at the beginning of 0.5 h and 1 h of MC3T3-E1 osteogenic differentiation.However,this phenomenon was significantly inhibited after the addition of recombinant human FGF18.Conclusion FGF18 promotes the proliferation but inhibits the differentiation and mineralization of pre-osteoblasts in vitro in a dose-de-pendent way within limits.Combined with the results of previous studies it is speculated that FGF18 may have a stage-spe-cific role in promoting/inhibiting osteogenic differentiation.FGF18 inhibits p-FOXO1 level at the beginning of pre-osteo-blasts differentiation and may thus mediate the inhibition of pre-osteoblast differentiation through this pathway.
