牛脂肪细胞分化过程中差异miRNA的鉴定与筛选
Identification and Screening of Differentially Expressed miRNAs During Bovine Adipocyte Differentiation

Abstract
This study aimed to analyze the differential expression of miRNAs in bovine adipocytes before and after differentiation and to elucidate their expression patterns and regulatory mechanisms.Bovine adipocyte samples collected at day 0 and day 6 of dif-ferentiation were subjected to high-throughput sequencing.Raw reads were preprocessed,and differentially expressed miRNAs were identified.Potential target genes of these miRNAs were then predicted and functionally annotated using bioinformatics tools.Finally,six miRNAs showing significant differential expression were selected for validation by qRT-PCR.The sequencing results yielded 701 known and 127 novel miRNAs across the three day 0 and three day 6 samples.Of these,668 miRNAs were expressed both before and after differentiation,while 160 miRNAs were uniquely expressed at one time point.Twenty-eight miRNAs were upregulated and nine were downregulated after differentiation.Gene Ontology enrichment analysis of the predicted targets revealed significant enrichment in biological processes such as protein binding and cytoskeletal organization,cellular components related to the actin cytoskeleton,and molecular functions including actin binding.KEGG pathway analysis showed that target genes were predominantly enriched in the MAPK,insulin,and adipokine signaling pathways;notably,the MAPK pathway plays key roles in cell proliferation,differentiation,and metabolism.qRT-PCR validation of the six selected miRNAs confirmed the sequencing trends.This study identified 37 differentially expressed miRNAs in bovine adipocytes before and after differentiation.Enrichment analysis revealed that their target genes are concentrated in signaling pathways such as MAPK,and subsequent validation experiments confirmed the reliability of the results.关键词
