木薯抗病蛋白RXam1互作蛋白的筛选与验证
Screening and Validation of RXam1 Interaction Protein of Manihot esculenta Disease Resistance Protein

Abstract
RXam1(ResistancetoXam1,RXam1)is a gene encoding the receptor-like kinase RLK.Its overex-pression can enhance resistance of Manihot esculenta to cassava bacterial wilt disease.To understand the regu-latory network of RXam1 in Manihot esculenta disease resistance,bioinformatics analysis revealed that MeRXam1 was a stable hydrophobic protein,containing two transmembrane domains and one signal peptide.There were 24 phosphorylation sites with a score higher than 0.8.MeRXam1 was expressed in eight tissues of Manihot esculenta,including leaves,petioles,stems,fragile embryonic callus,root tip meristem,fibrous roots,lateral buds and histalized embryonic structures.Phylogenetic tree analysis revealed that MeRXam1 had the highest protein homology with rubber trees.Secondly,the MeRXam1 gene was cloned into yeast-re-lated expression vectors and co-transformed into yeast AH109 cells with the Manihot esculenta pGBKT7 library.The interacting proteins of MeRXam1 were screened by yeast two-hybrid technology,and a total of 10 positive clones were obtained and sent for sequencing to further identify the interacting proteins.Among them,the interacting protein MeCRF5 encoded the RAP2.4 protein of the DREB subfamily in the AP2/ERF transcription factor superfamily,and the interaction between MeRXam1 and MeCRF5 was verified through yeast two-hybrid technology and bilayer fluorescence complementarity experiments.关键词
