基于SNP标记的八角遗传特征及性状关联分析
Genetic Characteristics and Trait Association Analysis of Illicium verum Based on SNP Markers

Abstract
Illicium verum is an important economic tree species that could be used both as medicine and food.The use of superior germplasm resources of Illicium verum for hybrid breeding had gradually become a new research topic in the next stage.However,the unclear genetic background of superior germplasm and the lack of molecular markers related to traits had hindered the breeding process.Based on the transcriptome data of Illicium verum,15 472 high-quality SNP loci were obtained.Through KASP typing identification,61 SNP loci with ideal typing effects were identified.Subsequently,genetic diversity statistical analysis and genetic structure analysis were conducted on 63 superior Illicium verum individual plants from 8 geographical popula-tions.The results indicated that the genetic diversity level of the peak group was relatively high among the eight groups.Except for the Rong'an group,there was a certain degree of inbreeding in most groups.The results of genetic structure analysis showed that the optimal number of subgroups was 4.Cluster analysis revealed that the octagonal population was not completely grouped into one category based on geographical origin,but was divided into 4 categories.Among them,the genetic information of the samples of category Ⅰ,Ⅱ and Ⅲ came from Subgroup 3,Subgroup 4,and Subgroup 1 respectively,while the sample of category Ⅳ had both the ancestors of Subgroup 2 and Subgroup 4.Further association analysis was conducted on SNP loci and flower color traits,and four SNP loci significantly associated with flower color traits were identified.Among them,phenotypic contribution rate was the highest,reaching 27.69%.Based on SNP loci,candidate genes significantly related to flower color traits,namely Glycogenphosphorylase1,beta-1,3-galactosyltrans-ferase2,Flavonoid3' hydroxylase and ANTHOCYANINLESS2,were identified.The genetic relationship and genetic structure among 63 superior individual Illicium verum plants were revealed through SNP mark-ers,and selected genes significantly related to flower color traits were obtained.This was conducive to more rational use of germplasm resources for breeding research.关键词
