Application and Optimization of A Rapid Detection Kit for Sulfur Dioxide in Traditional Chinese Medicine Analysis

Abstract
Objective:To establish a rapid screening method for sulfur dioxide(SO2)residues in traditional Chinese medicines(TCM).Methods:Six commercially available rapid detection kits for SO2 were evaluated for detection limits,linearity,operational convenience,and colorimetric performance,and one kit was selected for optimization.The sample pretreatment method and color card were optimized,and a mathematical model correlating the red(R),green(G),and blue(B)values with SO2 concentration was established employing Matlab 23.2.0.The optimized method was validated against the methods in the 2020 edition of the Chinese Pharmacopoeia.The SO2 residue in 251 batches of samples was determined,and the consistency of the results was assessed by the Kappa test.Results:The optimized kit demonstrated a detection limit of 30 mg·kg-1 with good stability and linearity.The sample pretreatment was simple and rapid.A standard color card with a concentration range of 30-600 mg·kg-1 was developed,and a mathematical model correlating R,G,B values with SO2 concentration was established.The quadratic polynomial fitting yielded coefficients of determination(R2)greater than 0.95 for the R,G,and B channels.The Kappa test indicated that the test results of the rapid detection kit were highly consistent with the method in the 2020 edition of the Chinese Pharmacopoeia(Kappa=0.911,P<0.001).Conclusion:The optimized rapid detection kit for SO2 can effectively detect SO2 residues in traditional Chinese medicine,providing an efficient and convenient method for detecting the residues of SO2 in TCM.关键词
