Abstract

Objective:To establish a fingerprint and a quantitative analysis of multi-components by single marker(QAMS)for the simultaneous determination of six components in Mongolian medicine Rhododendron micranthum and evaluate its quality.Method:An Agilent Eclipse Plus C18 column was used with a mobile phase consisting of 0.2%phosphoric acid solution(A)-acetonitrile(B)-tetrahydrofuran(C)with gradient elution(0-70 min,5%-20%B,2%-7%C)at a detection wavelength of 290 nm,so as to establish the high performance liquid chromatography(HPLC)fingerprints of seven batches of Rhododendron micranthum.Hyperoside was selected as the internal reference substance to develop the QAMS method for quantifying the other five components,and their contents were calculated using relative correction factors.The results were compared with those obtained by the external standard method(ESM)to evaluate the accuracy of QAMS.Results:The HPLC fingerprints revealed 20 common peaks in the seven batches of Rhododendron micranthum,with gallic acid,chlorogenic acid,hyperoside,isoquercitrin,avicularin,and quercitrin being assigned.The similarity between the sample fingerprints and the reference fingerprint ranged from 0.906 to 0.992.The relative standard deviation(RSD)of the above six components measured by QAMS and ESM was less than 3.0%.The content ranges of the six components were 0.083-0.121,0.041-0.206,2.422-7.844,1.934-4.234,0.334-2.477,and 3.205-6.424 mg·g-1,respectively.Conclusion:The HPLC fingerprint combined with the QAMS method is simple,accurate,and reproducible,providing a reliable approach for improving the quality control standards of Rhododendron micranthum.
