外源NO对酸胁迫下夏枯草种子萌发和幼苗生理的影响
Effects of exogenous nitric oxide on seed germination and seedling growth of Prunella vulgaris under acid stress

Abstract
[Objective]This study aimed to investigate the effects of exogenous nitric oxide(NO)on seed germi-nation and seedling physiology of Prunella vulgaris under acid stress.[Method]Seeds of P.vulgaris were soaked in sodium nitroprusside(SNP)solutions at different concentrations(0.01,0.05,0.1,0.5 and 1 mmol/L),using SNP solution as an exogenous nitric oxide donor.The seeds were then subjected to acid stress(pH=3.0).Germination parameters and seedling physiological indices were measured to evaluate the mitigating effects of exogenous NO un-der acid stress.[Results]Acid stress significantly inhibited seed germination,decreased photosynthetic pigment con-tent,and increased relative conductivity in P.vulgaris.In contrast,different concentrations of SNP effectively allevi-ated the acid-induced damage.The mitigation effect increased initiallyand then declinedwith rising SNP concentra-tion,with 0.1 mmol/L SNP showing the most pronounced improvements.Compared with the acid-stress control,the germination rate,germination potential,germination index,and vigor index increased by 36.36%,45.30%,46.32%and 75.83%respectively,while the germination recovery rate reached 28.57%.Root length,seedling height,fresh weight,and total chlorophyll content increased by 59.42%,23.21%,19.12%,and 19.27%,respec-tively,and relative conductivity decreased by 28.78%.Principal component analysis indicated that the effects of exog-enous NO on seed germination ability of P.vulgaris,under acid stress,were as follows:0.1 mmol/L SNP>0.5 mmol/L SNP>1 mmol/L SNP>0.05 mmol/L SNP>0.01 mmol/L SNP.[Conclusion]An appropriate con-centration of exogenous NO(0.1 mmol/L SNP)significantly enhances seed germination rate and seedling vigor of P.vulgaris under acid stress by reducing membrane damage and promoting photosynthetic chlorophyll accumulation.These findings provide a theoretical basis and technical reference for the ecological restoration and lands capeculiva-tionof P.vulgaris in acidified soils.关键词
