Protective effect of artemether on radiation-induced lung injury in nude mice bearing colorectal cancer

Abstract
Objective:Colorectal cancer is one of the most common malignancies worldwide,characterized by high incidence and mortality.Radiotherapy is a key therapeutic modality.Previous studies have suggested that artemisinin derivatives possess protective effects against radiation-induced injury.This study investigates the protective effect of artemether on radiation-induced lung injury in nude mice with colorectal cancer. Methods:An HCT116 colorectal cancer xenograft model was established in BALB/c nude mice by subcutaneous inoculation.Mice were randomly divided into 4 groups(n=5 in each group):a control group(intraperitoneal saline),a artemether group[intraperitoneal artemether at 50 mg/(kg·d)for 21 consecutive days],a radiotherapy group(intraperitoneal saline+local tumor X-ray irradiation at 2 Gy per fraction,every other day for a total of 5 fractions),and a combination group(artemether+radiotherapy using the same dosing schedule).Mice in the radiotherapy and combination groups received irradiation 1 hour after injection to induce radiation-induced lung injury.After the interventions,a blinded evaluator assessed general condition,body weight,organ indices,and lung injury. Results:Artemether improved general condition,activity level,and food/water intake in tumor-bearing mice with radiation-induced lung injury.There were no significant differences in body weight or liver index among the groups(all P>0.05).The spleen index in the radiotherapy group(0.004±0.001)was lower than that in the control group(0.005±0.002)and the artemether group(0.007±0.001)(both P<0.05),whereas the combination group(0.006±0.002)showed a higher spleen index than the radiotherapy group(P<0.05).The lung index in the radiotherapy group(0.014±0.002)was higher than that in the control group(0.009±0.003)and the artemether group(0.008±0.001)(P<0.05),while the combination group(0.011±0.003)had a lower lung index than the radiotherapy group(P<0.05).Histopathology showed that inflammatory cell infiltration,alveolar wall thickening,and interalveolar septal widening were alleviated in the combination group compared with the radiotherapy group.Lung injury scores in the radiotherapy group[3.00(3.00,4.00)]were higher than those of the control group[0.00(0.00,1.00)]and the artemether group[0.00(0.00,1.00)](both P<0.001),while the combination group[2.00(2.00,2.00)]had lower scores than the radiotherapy group(P=0.016,false discovery rate-adjusted). Conclusion:Artemether ameliorates acute radiation-induced inflammatory responses in lung tissue of colorectal cancer-bearing nude mice and enhances immune function,exerting a protective effect against radiation-induced lung injury.关键词
