Abstract

Objective:Human papillomavirus(HPV)is closely associated with the onset and progression of multiple diseases.Determining HPV infection status is of great importance for the diagnosis and treatment of various conditions.This study selects biopsy specimens from patients with HPV-positive cervical cytology to explore optimal methods for detecting HPV in paraffin-embedded tissue samples. Methods:Thirty paraffin-embedded cervical biopsy specimens were collected from patients with HPV-positive cervical cytology who underwent cervical biopsy during the same period at the Department of Pathology,Yichang Central People's Hospital,from January 2024 to December 2024.All cases showed positive cyclin-dependent kinase inhibitor 2A(CDKN2A,or p16)immunohistochemical staining.The optimal method for detecting HPV in paraffin-embedded tissue samples was identified by comparing different detection techniques(flow fluorescence analysis and hybridization chip assay),different DNA extraction methods(magnetic bead-based extraction and column-based extraction),and different quantities of tissue sections(6,8,10,and 12 6-μm paraffin sections). Results:Using 8 6-μm paraffin sections and extracting DNA with a column-based method,followed by PCR amplification and flow fluorescence HPV detection and genotyping,yielded the highest detection sensitivity and internal control qualification rates.The internal control qualification rate was 90%(27/30),and the detection sensitivity was 87%(26/30). Conclusion:The HPV detection method established in this study for paraffin-embedded tissue samples is simple to perform,allows for precise genotyping,and provides objective results without the need for manual interpretation.The method demonstrates high accuracy and good stability,making it suitable for widespread application in the pathological diagnosis and molecular typing of HPV-related diseases.It offers reliable support for clinical decision-making and prognostic assessment and holds significant clinical value.
