Effects of Exogenous Cytokinin Substances on Bulblet Propagation of Lycoris aurea

Abstract
[Objective]The effect of exogenous cytokinin substances on vegetative reproduction of Lycoris aurea was explored to provide the theoretical basis for improving vegetative propagation of L.aurea bulblets.[Method]The bulbet occurrence number,bulblet occurrence rate,bulblet propagation coefficient,root occurrence and surface morphological change of L.aurea bulbs treated with three concentrations(5 mg/L,25 mg/L and 50 mg/L)of 6-BA and three concentrations(150 mg/L,300 mg/L and 450 mg/L)of LOV respectively were compared by taking distilled water soaking treatment as CK to investigate the effect of 6-BA and LOV on bulblet propagation of L.aurea.[Result]The bulblet propagation coefficient showed an increasing trend with increase of 6-BA concentration when 6-BA concentration was 5-50 mg/L.The bulblet propagation coefficient of 6-BA1(5 mg/L)and 6-BA2(25 mg/L)treatments increased by 29.98%and 43.95%compared with CK.The bulblet propagation coefficient of 6-BA3(50 mg/L)treatment was 9.68,80.26%higher than CK.The bulblet propagation coefficient showed a declining trend with increase of LOV concentration when LOV concentration was 150-450 mg/L.The bulblet propagation coefficient of LOV3(450 mg/L)treatment was the minimum(3.45),35.75%lower than CK.There was no significant difference in the bulblet propagation coefficient between LOV treatments and CK.The optimal 6-BA concentration to promote bulblet vegetative propagation of L.aurea was 50 mg/L.The optimal LOV concentration to inhibit bulblet propagation was 450 mg/L.The bulblet formation days of 6-BA3,LOV3 and CK was 13 d,17 d and 15 d respectively.The bulblet occurrence number of 6-BA3 treatment after 45 d culture reached 277,25.91%higher than CK.The bulblet occurrence number of LOV3 treatment after 45 d culture was 158,28.18%lower than CK.The bulblet propagation coefficient of 6-BA3 and LOV3 treatment after 45 d culture was 3.30 and 1.88 respectively.The bulblet propagation coefficient of LOV3 treatment was lower than CK.The average occurrence rate of CK,6-BA3 and LOV3 treatments was 83.3%,91.7%and 67.8%separately.The average root length and root number of 6-BA3 and LOV3 treatment both were lower than CK.The 6-BA3 and LOV3 both could cause browning scale of L.aurea bulbs from the outer layer to the inner layer and the scale of the inner layer become curling.The scale curling and browning degree of 6-BA3 and LOV3 treatments was obviously deepened with increase of culture days.[Conclusion]6-BA has the promotion effect on bulblet propagation of L.aurea and inhibition effect on bulblet rooting of L.aurea.LOV has the inhibition effects on bulblet propagation and rooting of L.aurea.6-BA and LOV both can cause browning and curling of L.aurea bulbs.关键词
