新疆南疆地区犊牛冠状病毒的分离与鉴定及N基因进化分析
Isolation and Identification of Calf Coronavirus and Evolution Analysis of N gene in Southern Xinjiang

Abstract
Diarrhea cases of newborn calves across several large-scale dairy farms in southern Xinjiang prompted an investigation into the cause of calf diarrhea and to enrich the molecular epidemiology database for these farms.Viral isolation was performed from fecal samples of affected calves,followed by total RNA extraction.RT-PCR analysis and sequence alignment were conducted to evaluate homology of the isolated strains,and phylogenetic trees were constructed.The results showed that 9 out of 74 diarrhea samples were tested positive,the total positive rate was 12.2%.When the isolate was inoculated into human ileocecal carcinoma cells(HCT-8)and cultured through four generations,the cells exhibited typical pathological changes:rounding,shrinkage,stringing,ultimately obtaining the NJ-1 isolate.RT-PCR amplification revealed a 730 bp N gene,with genetic evolution analysis confirming 100.0%nucleotide homology to BCV-Aks-01 strain,indicating coalescence within the same lineage.These findings provide critical references for epidemiological studies of bovine coronavirus(BCoV)in southern Xinjiang's dairy farms.关键词
