Abstract
To explore the mechanism by which Mycobacterium intracellulare infection of mouse macrophage J774A.1 induces the secretion of IL-1β and IL-18,an infection model was established by infecting mouse macrophages with Mycobacterium intracellulare,while the control group cells were treated with an equal volume of sterile PBS.The concentrations of interleukin-1β(IL-1β)and interleukin-18(IL-18)were detected by ELISA,and the expression levels of NOD-like receptor pyrin domain-containing protein 3(NLRP3)and absent in melanoma 2(AIM2)mRNA were detected by real-time fluorescence quantitative PCR.After silencing the NLRP3 and AIM2 genes using small interfering RNA(siRNA)technology,the secretion levels of IL-1β and IL-18 were detected by ELISA.The results showed that compared with the control group,the secretion levels of IL-1β and IL-18 in the infection group were significantly increased(P<0.001),and the expression levels of NLRP3 and AIM2 mRNA were significantly increased(P<0.001).After silencing the NLRP3 and AIM2 genes,the secretion levels of IL-1β and IL-18 were significantly decreased(P<0.05).These results indicates that the secretion of IL-1β and IL-18 induced by Mycobacterium intracellulare infection of mouse macrophages depends on NLRP3 and AIM2 inflammasomes,which provides a reference for further exploration of the immune mechanism caused by Mycobacterium intracellulare infection in vivo.关键词
