Abstract

To compare the efficacy of magnetic bead-based and column-based method for extracting genomic DNA from Moschus grains,DNA was extracted from natural musk grains of varying masses using respective commercial kits.The quality concentration,purity,and integrity of the DNA obtained by both methods were compared using agarose gel electrophoresis and an ultra-micro nucleic acid analyzer.The utility of the extracted DNA was further assessed via conventional PCR and real-time fluorescent PCR.The results demonstrated that both methods successfully extracted genomic DNA even from the smallest sample mass(30 mg),and the DNA was suitable for PCR amplification of small target gene fragments.When the sample mass was≥50 mg,the DNA quality obtained by the magnetic bead-based method was slightly superior to that obtained by the column-based method.This study indicates that both extraction methods are capable of isolating DNA from natural musk grains,and the extracted DNA is suitable for detecting forest musk deer(Moschus berezovskii)-derived components using real-time fluorescent PCR.The selection of method can be made based on practical considerations.
