MCC950抑制人工关节磨损颗粒诱导炎性骨溶解的研究
Effect of MCC950 on wear particle-induced inflammatory osteolysis in artificial joints

Abstract
Objective To explore the effects of MCC950 on wear particle-induced osteolysis and inflammatory responses.Methods A total of thirty 8-week-old male C57BL/6 mice were selected as research subjects and randomly assigned into three groups using a random number table method:the sham group,the osteolysis model group,and the MCC950 intervention group,with 10 mice in each group.The sham group underwent skin incision and suturing only,while the other two groups were implanted with wear particles to establish an osteolysis model.The MCC950 intervention group received intraperitoneal injections of 5 mg/kg MCC950 every other day post-modeling,whereas the other two groups received equal-volume saline.After 14 days of intervention,serum levels of interleukin-1 β(IL-1β)and interleukin-18(IL-18)were measured.Calvarial specimens were collected for Micro-CT scanning and histological analysis.Results The bone mineral density,bone volume fraction,and bone area in the osteolysis model group were lower than those of the sham group,and the differences were statistically significant(P<0.05).The bone mineral density,bone volume fraction,and bone area in the MCC950 intervention group were higher than those of the osteolysis model group,and the differences were statistically significant(P<0.05).The porosity in the osteolysis model group was higher than that of the sham group,and the difference was statistically significant(P<0.05).The porosity in the MCC950 intervention group was lower than that of the osteolysis model group,and the difference was statistically significant(P<0.05).The osteoclast number in the osteolysis model group was higher than that of the sham group,and the difference was statistically significant(P<0.05).The osteoclast number in the MCC950 intervention group was lower than that of the osteolysis model group,and the difference was statistically significant(P<0.05).The serum levels of IL-1β and IL-18 in the MCC950 intervention group were lower than those of the osteolysis model group,and the differences were statistically significant(P<0.05).The serum levels of IL-1β and IL-18 in the osteolysis model group were higher than those of the sham group,and the differences were statistically significant(P<0.05).Conclusion MCC950 effectively suppresses wear particle-induced osteolysis and inflam-matory cytokine expression.关键词
