Construction of CDH11 Gene Overexpression Vector and Its Effect on Proliferation and Differentiation of Bovine Myoblasts

Abstract
The role of cadherin-11(CDH11)gene in the proliferation and differentiation of bovine adult myoblasts was investigated by overexpressing CDH11 and using EdU,CCK-8,immunofluores-cence and quantitative real-time PCR(qRT-PCR).The results showed that CDH11 overexpression significantly increased the EdU positivity and absorbance at 450 nm in bovine myogenic cells(P<0.05).The expression levels of cyclin-dependent kinase(CDK2)and Proliferating Cell Nuclear Anti-gen(PCNA)were significantly upregulated(P<0.05).During the myoblast differentiation stage,CDH11 overexpression significantly reduced the area of myotubes and markedly inhibited the myogen-ic differentiation-related genes myogenic differentiation(MYOD),myogenin(MYOG)and myogenic regulatory factors5(MYF5)expression(P<0.05).These findings indicate that CDH11 overexpres-sion promotes the proliferation of bovine adult myoblasts but inhibits the differentiation of bovine adult myoblasts.关键词
