马铃薯StCYP83B1基因RNAi遗传转化及功能分析

Abstract
Cytochrome P450(CYP450)is aheme-containing enzyme belonging to the B-family of the cytochrome P450 superfamily that plays a critical role in plant responses to biotic or abiotic stresses.Previous studies have identified a CYP450 gene,StCYP83B1 in potato,which is significantly induced upon infection by Phytophthora infestans;its overexpression enhances pattern-triggered immunity(PTI)and resistance to P.infestans without a growth penalty in potatoes.To further elucidate the role of StCYP83B1 in coordinating potato immunity and growth,this study generated StCYP83B1-RNAi transgenic potatoes via Agrobacterium-mediated genetic transformation.Comparative analyses with control plants('Atlantic')in immune response validation and growth parameter measurements demonstrated that StCYP83B1-RNAi plants exhibited reduced resistance to P.infestans,significantly down-regulated expression of PTI marker genes,and attenuated the flg22-induced reactive oxygen species(ROS)burst.Additionally,the StCYP83B1-RNAi plants showed significant decreases in plant height,leaf number,and stem diameter.These findings preliminarily confirm that StCYP83B1 posi-tively regulates disease resistance and is essential for maintaining normal growth in potato.This study not only provides a theoretical reference for molecular resistance breeding,but also provides critical material for further mechanistic investigation of how StCYP83B1 coordinates potato immunity and growth.关键词
