Abstract

This study aimed to investigate the structure and compositional differences in the seed coat of Cucurbita moschata cultivars with different seed coat thicknesses and to elucidate the relationship among seed coat thickness,seed coat structure and component accumulation,laying a foundation for understanding the mechanism of thin seed coat formation and to provide theoretical guidance for the breeding hull-less C.moschata varieties.One normal-seeded and seven thin-coated C.moschata culti-vars with different seed coat thicknesses were used as experimental materials.At 50 days after pollina-tion,seed phenotypes and seed coat microstructures were observed using scanning electron microsco-py,and the contents of lignin,cellulose,and starch in the seed coats were quantified.The results showed that the thinner the seed coat,the greener the seed appearance.The reduction in seed coat thickness in thin-coated cultivars was mainly due to the collapse of three tissue layers:the hypodermis(H),parenchyma(A),and chlorenchyma(C).The contents of lignin,cellulose,and starch were posi-tively correlated with seed coat thickness;the lower their content,the thinner the seed coat.Decreased accumulation or deposition of lignin,cellulose,and starch led to the collapse of the H,A,and C layers,ultimately resulting in the formation of thin seed coats.As the outer seed coat materials diminished,the innermost chlorenchyma(C)layer became more visible,giving the seeds a greener appearance.
