祁连山野生鳞伞的鉴定、生物学特性与驯化栽培
Identification,Biological Characteristics and Domestication of Wild Pholiota Species in Qilian Mountains

Abstract
Toexploit wild edible mushroom resources in the Qilian Mountains and facilitate their do-mestication,strain Pa-01 was isolated from wild Pholiota fruiting bodies collected in the Gansu Qilian Mountain National Nature Reserve.Strain Pa-01 was identified as Pholiota adiposa(Batsch)P.Kumm.through morphological observation and ITS sequence analysis.The effects of temperature,pH,carbon and nitrogen sources,carbon-to-nitrogen ratio(C/N),inorganic salts,and vitamins on my-celial growth were systematically investigated.Results showed optimal growth at 22 ℃ and pH 6.5,with fructose as the preferred carbon source,yeast extract as the optimal nitrogen source,and a C/N ratio of 20∶1.Supplementation with MnSO4 and vitamin B12(VB12)significantly enhanced mycelial biomass accumulation.Orthogonal experiments revealed the optimal culture conditions(A3B1C3D3)as follows:mannitol(carbon source),yeast extract(nitrogen source),MnSO4 supplementation,and incubation at 26 ℃.Cultivation trials utilizing locally sourced substrates achieved a biological efficien-cy of 110%for fruiting bodies using a mixture of corn cob(83%),wheat bran(15%),and lime(2%).This study provides the first comprehensive analysis of the biological traits and cold-adaptive mechanisms of P.adiposa in the Qilian Mountains.The corn cob-based,cost-effective cultivation protocol offers a sustainable strategy for wild germplasm conservation and sustainable utilization.关键词
