Abstract

The effects of the volume of chromogenic solution,chromogenic temperature,and chromogenic time on the absorbance values were investigated separately using anthrone/phenol-vitriolic colorimetry for the determination of polysaccharides in Dendrobium devonianum.Response surface methodology was applied to model and optimize the factors influencing the absorbance of polysaccharides in artificially greenhouse-cultivated D.devonianum.Experimental results showed that in the determination of polysaccharides in artificially greenhouse-cultivated D.devonianum using the anthrone-sulfuric acid method,chromogenic time had the greatest effect on the absorbance,followed by the amount of anthrone solution,and finally the chromogenic temperature.In contrast,the application of the phenol-sulfuric acid method revealed that the volume of phenol solution exerted the greatest influence on the absorbance,followed by the chromogenic time,while the chromogenic temperature demonstrated the least effect.Both methods produced well-fitted standard curves with high precision,good stability,and high recovery rates under optimized conditions,confirming their suitability for determining polysaccharide concentrations in artificially greenhouse-cultivated D.devonianum.Significant discrepancies were observed in the experimental data with the application of these two methods for determining polysaccharide concentrations in D.officinale and D.devonianum,respectively.
