The NOX4/ROS/GSDMD axis in curcumin-induced pyroptosis and growth inhibition of HepG2 cells

Abstract

The study explores the effects and molecular mechanisms of curcumin(Cur)on pyroptosis in human hepatocellular liver carcinoma cells(HepG2).The effects of curcumin on cell viability,pyroptosis,apoptosis,and oxidative stress in HepG2 cells,along with its regulatory mechanism on pyroptosis,were investigated using CCK-8 assay,Annexin V-FITC/PI double staining,DCFH-DA fluorescent probing,Western blot analysis,and enzyme-linked immunosorbent assay(ELISA).The results showed that the viability of HepG2 cells was significantly inhibited by curcumin in a dose-dependent manner;curcumin-treated HepG2 cells displayed abnormal morphology with many vacuoles,and there was a significant increase in LDH,IL-1β,and IL-18.Intracellular levels of ROS and MDA in HepG2 cells were significantly up-regulated by curcumin in a dose-dependent manner.Furthermore,curcumin notably promoted the phosphorylation of NF-κB in HepG2 cells as well as increased the expression levels of NLRP3,NOX4,caspase-1,and gasdermin D(GSDMD)proteins.The results suggest that curcumin may induce pyroptosis in HepG2 cells by activating the NOX4/ROS/GSDMD axis,promoting NLRP3 inflammasome assembly,which subsequently activates Caspase-1 and cleaves GSDMD,leading to the release of LDH,IL-1β,and IL-18.
