OCIAD2在胰腺癌中的高表达及其生物学功能与临床意义分析
Biological Function and Clinical Significance of Overexpressed OCIAD2 in Pancreatic Cancer

Abstract
Objective To investigate the expression,biological function,and clinical significance of OCIAD2 in pancreatic cancer.Methods The differential expression of OCIAD2 between pancreatic cancer and normal tissues was analyzed by GE-PIA2 software based on the TCGA and GTEx databases,which was further validated using 3 pairs of pancreatic cancer and adjacent non-tumor tissues.With the TCGA pancreatic adenocarcinoma(PAAD)dataset,Pearson correlation analysis was performed to assess the correlation between OCIAD2 expression and methylation of specific CpG sites.Weighted Gene Co-expression Network Analysis(WGCNA)and Gene Ontology(GO)enrichment analysis were conducted to explore the bio-logical functions of OCIAD2.Single-sample Gene Set Enrichment Analysis(ssGSEA)was used to evaluate the correlation between OCIAD2 expression and immune cell infiltration.GEPIA2 software was employed to analyze the impact of OCIAD2 expression and methylation of its cg26134090 site on overall survival(OS)and disease-free survival(DFS)of patients.Additionally,the receiver operating characteristic(ROC)curves were plotted based on the GSE28735,GSE62452,and GSE16515 datasets using the R language pROC package to evaluate the diagnostic efficacy of OCIAD2.Results The OCIAD2 expression was higher in pancreatic cancer tissues than in normal tissues(P<0.01),and its expression was nega-tively correlated with the methylation degree of cg26134090 site(r=-0.52,P<0.001).WGCNA and GO enrichment analy-ses revealed that gene sets for cell adhesion(GO:0007155)and negative regulation of cell proliferation(GO:0008285)were significantly enriched in the low OCIAD2 expression group.Immune infiltration analysis indicated a negative correla-tion between the infiltration of effector memory CD4+T cells and OCIAD2 expression(r=-0.501,P<0.001).ROC curves based on the GSE28735,GSE62452,and GSE16515 datasets demonstrated that the areas under the curve(AUC)for OCIAD2 in diagnosing pancreatic cancer were 0.872,0.853,and 0.818,respectively.Survival analysis showed that pa-tients with high OCIAD2 expression had shorter OS and DFS(OS:HR=2.4,P<0.01;DFS:HR=2.2,P<0.05).Conclu-sion OCIAD2 is upregulated in pancreatic cancer,potentially associated with hypomethylation at its cg26134090 site.Bioinformatics suggests that OCIAD2 plays a role in pancreatic cancer progression possibly by regulating cell adhesion and proliferation,which is associated with the tumor immune microenvironment.OCIAD2 demonstrates excellent diagnostic val-ue for pancreatic cancer,and its high expression indicates an unfavorable prognosis for patients.关键词
