Abstract

Objective To investigate the pro-oncogenic role of thrombospondin-1(THBS1)in gastric cancer and its under-lying biological mechanisms.Methods Clinical samples were analyzed to examine THBS1 expression in gastric cancer tis-sues and its correlation with patient prognosis.The effects of THBS1 overexpression or knockdown on the proliferation of gastric cancer cells were assessed using CCK-8 and colony formation assays.Wound healing and Transwell chamber assays were employed to evaluate the impact of THBS1 expression on cell migration and invasion.Transcriptome sequencing was performed to identify key signaling pathways regulated by THBS1.Western blot and nuclear-cytoplasmic fractionation were used to detect the expression and phosphorylation of Hippo pathway proteins,including MST1/2,LATS1/2,YAP1,and TEAD1.Results Clinical analysis revealed that THBS1 was upregulated in gastric cancer,which was associated with poor prognosis.In vitro experiments demonstrated that the knockdown of THBS1 significantly inhibited the proliferation,migra-tion,and invasion of gastric cancer cells AGS and SGC7901.Transcriptome sequencing,Western blot,and nuclear-cyto-plasmic fractionation indicated that the Hippo signaling pathway was markedly activated through suppressing the THBS1 ex-pression.Rescue experiments confirmed that pretreatment with XMU-MP-1 significantly reversed the inhibitory effects of THBS1 knockdown on cell proliferation and migration.Conclusion THBS1 promotes the malignant progression of gastric cancer cells by activating the Hippo signaling pathway.
