PDCD4 Contributes to Postoperative Delirium Through the AKT-BDNF Pathway

Abstract
Objective To explore the role of PDCD4 in postoperative delirium through the AKT-BDNF pathway.Methods The delirium-like behaviors were observed by constructing an anesthesia/surgery mouse model.Western blot was performed to assess the protein levels of PDCD4,AKT,p-AKT,p-CREB,and BDNF in the hippocampus of mice.Behavioral and protein changes were further verified using PDCD4 knockout mice and administering an overexpression virus.Results Be-havioral tests indicated that mice undergoing anesthesia/surgery exhibited delirium-like behaviors,with elevated PDCD4 in the hippocampus and inhibited AKT-BDNF pathway.PDCD4 knockout mice had improved delirium-like behaviors,along with activation of the AKT-BDNF pathway.Moreover,delirium-like behaviors were also observed in wild-type(WT)mice after the injection of the PDCD4 overexpression virus into the hippocampus.Conclusion PDCD4 is involved in postopera-tive delirium through the AKT-BDNF pathway.关键词
