Abstract

Objective To investigate the therapeutic effect of amniotic fluid-derived mesenchymal stem cells(AF-MSCs)in a mouse model of intrauterine adhesions(IUA),and to reveal their potential mechanism by combining bioinformatics.Methods An IUA model was established using mechanical scraping followed by treatment with 95%ethanol.Two weeks af-ter surgery,AF-MSCs were injected into the uterine cavity,and uterine tissues were collected after another two weeks.En-dometrial thickness was assessed via HE staining,and we compared the fibrotic area using Masson staining.The expressions of CK19 and Vimentin were detected by immunohistochemistry,while differentially expressed RNAs were identified through RNA sequencing.Results Treatment with AF-MSCs resulted in a reduction in the extent of IUA,an increase in endometri-al thickness,and a decrease in the degree of fibrosis.The mechanism may be related to the up-regulation of the expression of genes related to ovarian sex hormone secretion and cell proliferation.Conclusion AF-MSCs significantly promote the proliferation of endometrial cells and improve the fibrosis associated with IUA,which provides a theoretical basis for AF-MSC therapy in the treatment of IUA.
