Inferring Postmortem Interval Based on Changes in Amino Acids in Liver Tissue:A Preliminary Study

Abstract
Objective To detect the temporal variations in amino acids in postmortem liver tissue using gas chromatography-mass spectrometry(GC-MS)and to investigate the optimal equation for inferring the postmortem interval(PMI).Meth-ods A model simulating postmortem changes in isolated pig liver tissue was established,maintaining a constant temperature of 25℃ and humidity of 75%.Morphological assessments,including measurements of the total interstitial tissue area and total collagen fiber content,were conducted at 12-h intervals spanning from 0 h to 120 h PMI.Concurrently,samples were collected for amino acid extraction,and GC-MS was employed to analyze the correlation between changes in amino acids and PMI,ultimately establishing an optimal inference equation.Results The total interstitial tissue area,total collagen fi-ber content,and amino acid levels in isolated liver tissue exhibited temporal changes in relation to PMI.Notably,GC-MS revealed a strong correlation between alanine,leucine,valine,and PMI,yielding the inference equation:YPMI=11·456 Xleucine+13.605Xalanine+14.162Xglycine+6.110(R2=0.922,SE=10.629).Conclusion This study preliminarily validates the potential of using amino acids in liver tissue as a novel approach for inferring PMI during the early to middle stages of de-composition,suggesting promising application prospects.关键词
