Abstract

Objective:To investigate impact of tumor necrosis factor receptor-associated factor 2(TRAF2)on Mycobacterium smegmatis(M.smeg)infection in macrophages and elucidate its underlying mechanism.Methods:The TRKD cell line was estab-lished by lentiviral infection of RAW264.7 macrophages to achieve stable knockdown of TRAF2 protein expression,while the scramble cell line was used as a negative control.The impact of TRAF2 on macrophage phagocytosis and M.smeg killing was investigated using qRT-PCR,Western blot,immunofluorescence staining,flow cytometry,single colony counting and other experimental techniques to elucidate the underlying mechanism.Results:After infection with M.smeg,the phosphorylation of TRAF2 in RAW264.7 cells was sig-nificantly inhibited.Compared to scramble cells,TRKD cells exhibited prominent protruding tentacles,weakened phagocytosis and bactericidal activity against M.smeg,as well as suppressed apoptosis.Mechanistic studies revealed that compared to scramble cells,treatment with M.smeg or LPS inhibited the M1 polarization of TRKD cells while enhancing IL-4-induced M2 polarization;this phe-nomenon was associated with the inhibition of the NF-κB pathway but not the MAPK signaling pathway.Conclusion:Silencing TRAF2 can impede the M1 polarization of macrophages induced by M.smeg infection,thereby attenuating their phagocytic and bactericidal ca-pacity against M.smeg,underscoring the pivotal protective role played by TRAF2 in macrophage-mediated defense against M.smeg.
