摘要

Abstract

Objective:To investigate the effect of cyclicRNA VMA21(CircVMA21)on lipopolysaccharide(LPS)induced damage of alveolar epithelial cells by regulating the microRNA-122-5p(miR-122-5p)/nuclear factor Ⅸ(NFⅨ)axis.Methods:A549 cells were treated with 1 μg/ml LPS and separated into LPS group,pcDNA-NC group(pcDNA-NC),pcDNA-VMA21 group(transfected with pcDNA-VMA21),pcDNA-VMA21+miR-NC group(transfected with pcDNA-VMA21+miR-NC),and pcDNA-VMA21+miR-128 mimics group(transfected with pcDNA-VMA21+miR-128 mimics).A549 cells that did not undergo any treatment were recorded as NC group.qRT-PCR was applied to detect the expressions of CircVMA21 and miR-122-5p in A549 cells in each group;CCK-8 meth-od was applied to detect A549 cell viability;flow cytometry was applied to detect apoptosis in A549 cells;ELISA method was applied to detect the levels of TNF-α,IL-1β and IL-6 in A549 cells;Western blot was applied to detect levels of NFⅨ,B-cell lymphoma gene-2(Bcl-2),BCL2 associated X protein(Bax),activated cysteine asparase-3(cleaved-caspase-3),E-cadherin,N-cadherin and Vimentin proteins.Results:Compared with NC group,the CircVMA21 and NFⅨ levels,A450 value,E-cadherin and Bcl-2 levels in LPS group were significantly reduced(P<0.05),the apoptosis rate,miR-122-5p level,Bax,cleaved-Caspase-3,N-cadherin,Vimen-tin protein levels,TNF-α,IL-1β and IL-6 levels were significantly increased(P<0.05);compared with LPS group and pcDNA-NC group,the CircVMA21 and NFⅨ levels,A450 value,E-cadherin and Bcl-2 levels in pcDNA-VMA21 group were significantly in-creased(P<0.05),the apoptosis rate,TNF-α,IL-1β,and IL-6 levels,miR-122-5p level,Bax,cleaved-Caspase-3,N-cadherin,Vi-mentin protein levels were significantly reduced(P<0.05);overexpression of miR-122-5p reversed the improvement effect of upregula-tion of CircVMA21 expression on LPS-induced A549 cell damage;CircVMA21 targeted and regulated the miR-122-5p/NFⅨ axis.Conclusion:CircVMA21 may improve LPS-induced A549 cell damage by down-regulating miR-122-5p and up-regulating NFⅨ.
