南蛇藤素对心肌梗死大鼠心肌重塑的改善作用及有关机制
Improvement of celatrol on myocardial remodeling in rats with myocardial infarction and its related mechanism

Abstract
Objective:To investigate impact of celatrol on myocardial remodeling in rats with myocardial infarction(MI)and its mechanism.Methods:SD rats were randomly grouped into sham operation(Sham)group,MI group,cGAS inhibitor(RU.521)group,low,medium and high doses celatrol(CEL-L,M,H)groups,with 10 rats in each group.MI model was established by ligating anterior descending branch of left coronary artery.After administration,indexes of left ventricular function,level of serum inflammatory factors,pathological changes and fibrosis of myocardium,and expressions of genes and proteins related to cGAS-STING signal path-way in myocardial tissue were detected.Results:Compared with Sham group,left ventricular end-diastolic diameter(LVEDd),left ventricular end-systolic diameter(LVESd),left ventricular posterior wall thickness dimensions(LVPWd),left ventricular posterior wall thickness at end-systolic(LVPWs),IFN-β,interferon-induced protein 10(CXCL10),TNF-α levels and proportion of collagen fibers in myocardial tissue,mRNA and protein expressions of cGAS and STING,p-NF-κB p65/NF-κB p65 in MI group were increased,levels of left ventricular ejection fraction(LVEF)and left ventricular fraction shortening rate(LVFS)were decreased(P<0.05),and there was obvious pathological damage in myocardial tissue;compared with MI group,RU.521 group and CEL-L,M,H groups were able to obviously improve changes of above indexes in rats,and the higher the dose of celatrol,more obvious improvement of above indexes;RU.521 and CEL-H had almost same effect on above indicators.Conclusion:Celatrol can improve adverse myocardial remodeling in MI rats,whose mechanism may be related to inhibition of inflammation mediated by cGAS-STING signal pathway.关键词
