Elucidating localization and biological function of Pb29:A prospective candidate gene for malaria transmission-blocking vaccines

Abstract
Objective:To investigate the expression and subcellular localization of Pb29 in P.berghei,to clarify the function of Pb29 in the sexual development stage of P.berghei,and to provide experimental basis for the development of candidate antigens of transmission blocking vaccine.Methods:The homology and conservation of Pb29 were determined by bioinformatics analysis.The Pb29 gene knockout strain(Pb29-KO)and Pb29 tag strain(Pb29-HA)were obtained by double-cross homologous recombination,and the influence of Pb29 gene on the growth and development of P.berghei was analyzed by phenotypic experiment.The expression stage and localization of Pb29 protein during the development of P.berghei were detected by IFA and Western blot.Results:BLAST homolo-gy sequence analysis revealed that Pb29 exhibits high homology and conservation across various Plasmodium species,encoding a pro-tein possesses a signal peptide.Western blot and IFA showed that Pb29 was expressed in the schizont,gametocyte and ookinete stag-es,mainly on the surface of the ookinete membrane.Phenotypic experiments confirmed that Pb29 had no effect on the development of P.berghei in asexual blood stage,but inhibited the formation of ookinetes and oocysts in mosquito stomach.Conclusion:Pb29 is pre-dominantly expressed on the surface of the ookinete membrane.The knockout of Pb29 resulted in the inhibition of ookinetes and oo-cysts formation in the mosquito midgut,indicating its potential as a candidate gene for the development of transmission-blocking vac-cines.关键词
