紫花前胡素调节Notch-1/Hes-1信号通路对LPS诱导的关节软骨细胞损伤的影响
Effect of decursin on LPS induced joint chondrocyte injury by regulating Notch-1/Hes-1 signaling pathway

Abstract
Objective:To investigate effect of decursin(DE)on lipopolysaccharide(LPS)induced joint chondrocyte injury by regulating Notch-1/Hes-1 signaling pathway.Methods:ATDC5 cells were separated into control group,LPS group,low,medium,high-doses DE groups and high-dose DE+Jagged1 group.EdU staining and CCK-8 method were applied to detect ATDC5 cell prolifera-tion.Flow cytometry was applied to detect apoptosis of ATDC5 cells.ELISA was applied to detect levels of IL-6 and TNF-α in superna-tant of ATDC5 cells.qRT-PCR was applied to detect mRNA expressions of collagen type Ⅱ alpha 1(COL2α1),matrix metalloprotein-ase-13(MMP-13),Bcl2 associated X protein(Bax)and Caspase-3 in ATDC5 cells.Western blot was applied to detect expressions of Notch-1 and Hes-1 proteins in ATDC5 cells.Results:Compared with control group,EdU positive cell rate,A450 and COL2α1 mRNA expression in ATDC5 cells were reduced in LPS group,apoptosis rate,IL-6 and TNF-α levels in supernatant,MMP-13,Bax,Cas-pase-3 mRNA expressions,Notch-1 and Hes-1 protein expressions in ATDC5 cells were increased(P<0.05).Compared with LPS group,EdU positive cell rate,A450 and COL2α1 mRNA expression in ATDC5 cells were increased in low,medium and high-doses DE groups,apoptosis rate,IL-6 and TNF-α levels in supernatant,MMP-13,Bax,Caspase-3 mRNA expressions,Notch-1 and Hes-1 protein expressions in ATDC5 cells were reduced(P<0.05).Compared with high-dose DE group,EdU positive cell rate,A450 and COL2α1 mRNA expression in ATDC5 cells were reduced in high-dose DE+Jagged1 group,apoptosis rate,IL-6 and TNF-α levels in supernatant,MMP-13,Bax,Caspase-3 mRNA expressions,Notch-1 and Hes-1 protein expressions in ATDC5 cells were increased(P<0.05).Conclusion:DE may promote LPS-induced proliferation of ATDC5 cells,inhibit apoptosis,inflammation,and extracellu-lar matrix degradation by inhibiting Notch-1/Hes-1 pathway,thereby reducing cell damage.关键词
