银杏内酯A通过调控MAPK/STAT3信号轴改善小鼠急性胰腺炎
Ginkgolide A ameliorates acute pancreatitis in mice by regulating MAPK/STAT3 signaling axis

Abstract
Objective:To investigate effects of ginkgolide A on acute pancreatitis(AP)mice and to preliminarily explore potential mechanism.Methods:An AP mouse model was constructed by intraperitoneal injection of caerulein 50 μg/kg.Successfully modeled AP mice were randomly divided into model group,ginkgolide A group(750 mg/kg),anisomycin group(750 mg/kg ginkgolide A by gavage+5.0 mg/kg anisomycin by intraperitoneal injection)and Colivelin group(750 mg/kg ginkgolide A by gavage+0.5 mg/kg Co-livelin by intraperitoneal injection),with 8 mice in each group.Routinely cultured C57BL/6 mice were used as control group(n=8).After 24 h of drug treatment,pancreas was weighed and pancreatic index was calculated.Serum amylase,lipase,TNF-α,IL-1β,IL-8,IL-6,monocyte chemotactic protein(MCP)-1 and macrophage inflammatory protein(MIP)-1α levels were detected by ELISA.HE staining was used to observe pancreatic pathological tissue damage.Immunohistochemical was used to assess inflammatory cell in-filtration of pancreatic tissue.Western blot was used to analyze expressions of NF-κB,p38 MAPK,ERK and STAT3 pathway proteins.Results:Ginkgolide A reduced pancreatic index and serum levels of amylase,lipase,TNF-α,IL-1β,IL-8,IL-6,MCP-1 and MIP-1α in AP mice,attenuated pancreatic tissue pathological injury,reduced pancreatic tissue macrophage and neutrophil infiltration,and inhibited phosphorylation of p65,IκBα,p38 MAPK,ERK and STAT3.Anisomycin,a MAPK-specific activator,and Colivelin,an ERK-specific activator,were able to abrogate effects of ginkgolide A on pancreatic injury,inflammatory response and MAPK/STAT3 pathway activation in AP mice to a certain extent.Conclusion:Ginkgolide A improves caerulein-induced pancreatic function and pathological injury,whose mechanism maybe related to inhibition of MAPK/STAT3 signaling axis activation.关键词
