Effect of kaempferol regulating JAK2/STAT3 signaling pathway on inflam-matory response and hearing impairment in rats with secretory otitis media

Abstract

Objective:To investigate the impacts of kaempferol(Kae)on inflammatory response and hearing impairment in rats with secretory otitis media(SOM)by regulating the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods:A SOM rat model was established,the experiments were divided into Control group,SOM group,Kaonophenol low-,medium-and high-dose[Kae-L,Kae-M,Kae-H,0.35,0.70,1.40 mg/(kg·d)Kae]groups and high dose kaempferol+JAK2/STAT3 activator RO8191[Kae-H+RO8191,100 mg/(kg·d)Kae+21 mg/(kg·d)RO8191]group.Auditory brainstem evoked potential was applied to detect auditory function;levels of associated inflammatory factors were detected by ELISA;micrometers was applied to measure the thickness of ear mucosal tissue;HE and PAS staining were applied to observe pathological changes of ear mucosal tissue and proliferation of goblet cells;Western blot was applied to detect levels of JAK2/STAT3 signaling pathway related proteins.Results:Compared with Control group,SOM group showed cilia shedding,cell swelling,thickening of the middle ear mucosa and submucosal tissue,increased infiltration of inflammatory cells,and increased goblet cells;ABR threshold,IL-6,IL-4,IL-1β levels,eardrum thickness,JAK2,STAT3 phosphorylation levels of rats were obviously increased(P<0.05).Compared with SOM group,Kae-L,Kae-M and Kae-H groups showed decreased cilia shedding,cell swelling,thinning of middle ear mucosa and submucosal tissue,reduced infiltration of inflammatory cells,and decreased proportion of goblet cells;ABR threshold,IL-6,IL-4,IL-1β levels,eardrum thickness,JAK2,STAT3 phosphorylation levels in rats were decreased in a dose-dependent manner(P<0.05).RO8191 reversed the therapeutic effect of Kae on SOM rats(P<0.05).Conclusion:Kae may inhibit inflammatory response of SOM rats and alleviate hearing impairment by inhibiting JAK2/STAT3 signaling pathway.
