Abstract

Objective:To investigate protective effect of IL-35 against oxygen-glucose deprivation/reperfusion(OGD/R)-in-duced microglia BV2 injury and possible mechanism.Methods:BV2 cells were cultured in vitro,and experiment was divided into Control group,OGD/R group and IL-35 group;CCK-8 was used to detect cell viability,colorimetric assay was used to detect cellular oxidative stress level,ELISA was used to detect expressions of inflammatory factors,high-throughput sequencing was used to analyze differential genes,PCR and Western blot were used to detect expressions of cGAS and STING gene and protein.Results:Compared with OGD/R group,viability of BV2 cells in IL-35 group was significantly increased,expressions of ROS,MDA,IL-6,IL-1β and TNF-α in IL-35 group were significantly decreased,while expression of SOD was significantly increased.Differential gene analysis showed that between OGD/R group and IL-35 group,there were 290 up-regulated genes and 261 down-regulated genes;KEGG enrich-ment analysis showed that down-regulated genes were mainly enriched in cytoplasmic DNA sensing pathway.PCR and Western blot results showed that cGAS and STING gene and protein expressions in IL-35 group were significantly lower than OGD/R group(all P<0.05).Conclusion:IL-35 has a protective effect on OGD/R-induced BV2 cell damage,and its mechanism may be related to cGAS-STING signalling pathway.
