Effects of catalpol on proliferation,migration and immune escape of endometrial cancer cells by regulating CCL2-CCR2 axis

Abstract
Objective:To investigate the effects of catalpol on proliferation,migration,immune escape and CC chemokine ligand 2(CCL2)-CC chemokine receptor 2(CCR2)signal axis in endometrial cancer(EC)cells.Methods:EC cells Ishikawa were cultured and treated with 0,0.125,0.25,0.5,1.0 and 2.0 mmol/L catalpol.MTT method was applied to detect cell viability,and appropriate concentrations were selected for subsequent experiments.EC cells Ishikawa were grouped into Control group,low,medium and high concentration treatment groups(Catalpol-L,Catalpol-M,Catalpol-H groups),and high concentration treatment+CCL2 recom-binant protein group(Catalpol-H+CCL2 group);EdU experiment,plate cloning experiment,flow cytometry and scratch experiment were respectively applied to detect cell proliferation,apoptosis and migration abilities;Western blot was applied to detect expression levels of Ki67,Cyclin D1,Caspase-3,Bax,MMP-2,MMP-9,CCL2 and CCR2 proteins;Ishikawa cells were co-cultured with CD8+T cells,and trypan blue staining was applied to detect CD8+T cell viability;ELISA was applied to detect levels of PD-L1,IL-10,IL-6 and TGF-β.Results:0.25,0.5 and 1.0 mmol/L catalpol were selected to treat Ishikawa cells for subsequent experiments;compared with Control group,positive rate of EdU,colony formation,scratch healing rate,and expression levels of Ki67,Cyclin D1,MMP-2,MMP-9,CCL2 and CCR2 in Catalpol-L,Catalpol-M and Catalpol-H groups were obviously reduced,while apoptosis rate,Caspase-3 and Bax were obviously increased(P<0.05);compared with Catalpol-H group,positive rate of EdU,colony formation,scratch healing rate,and expression levels of Ki67,Cyclin D1,MMP-2,MMP-9,CCL2 and CCR2 in Catalpol-H+CCL2 group were obviously increased,while apoptosis rate,Caspase-3 and Bax were obviously reduced(P<0.05).Co-culture results of Ishikawa cells and CD8+T cells showed that compared with Control group,survival rate of CD8+T cells in Catalpol-L,Catalpol-M and Catalpol-H groups was obviously increased,while levels of PD-L1,IL-10,IL-6 and TGF-β were obviously decreased(P<0.05);compared with Catalpol-H group,survival rate of CD8+T cells in Catalpol-H+CCL2 group was obviously reduced,while levels of PD-L1,IL-10,IL-6 and TGF-β were obviously increased(P<0.05).Conclusion:Catalpol can inhibit EC cell proliferation,migration and immune escape,and promote EC cell apoptosis by inhibiting the CCL2-CCR2 axis.关键词
