Full-length transcriptome sequencing and characteristics analysis of vision-related functional genes in Odontodactylus scyllarus

Abstract
The genetic characteristics underlying the unique vision of stomatopods remain unexplored.To explore functional genes related to stomatopod vision and their structural characteristics,this study selected Odontodactylus scyllarus as the research object and performed full-length transcriptome sequencing for the first time.After filtering low-quality sequencing data,17105 Unigene sequences were obtained through assembly.Using a hidden Markov model,23 opsin genes were identified from Unigene sequences,comprising 17 long-wavelength-sensitive opsin genes(LWS),four medium-wavelength-sensitive opsin genes(MWS),and two ultraviolet-sensitive opsin genes(UV).The amino acid lengths,molecular weights,and isoelectric points of these 23 opsin genes ranged from 188 to 433 aa,21.04 to 48 kDa,and 5.89 to 9.20,respectively.They contained basic amino acids and exhibited high hydrophobicity and stability.Motif analysis revealed seven transmembrane domains in all 23 opsin genes,with the seventh transmembrane domain containing conserved QAKKMH and SLR motifs.In opsin secondary structure,the α-helix constitutes the largest proportion,followed by irregular coils,extended chains,and β-folds.The MWS subfamily showed the highest sequence identity(89.35%),followed by the UV subfamily(76.18%)and the LWS subfamily(72.07%).Additionally,functional genes associated with light conduction—including TRPL,RK,PKC,DAGL,Rh,PLCβ,NINAC,IP3R,Arr2,Actin,and CamKII were screened.The full-length transcriptome sequence of O.scyllarus will enrich genetic information databases for O.scyllarus and other stomatopods,laying the foundation for exploring the genetic mechanisms and evolutionary processes behind stomatopod special vision.关键词
