Abstract

This study employed environmental DNA(eDNA)metabarcoding technology with a local 12S rDNA database of Yellow and Bohai Sea fishes to monitor fish diversity in Jincheng Marine Ranch,Laizhou Bay.Key findings include:(1)84 fish species significantly surpassed the four species recorded during baseline surveys using gillnets,traps,and longlines in 2009 during ranch construction,demonstrating the superior sensitivity of eDNA technology and providing reference data for the diversity of fish;(2)41 species(48.81%of eDNA findings,50.62%of historical records)overlapped with historical Laizhou Bay records,while the remaining 43 species represent Yellow-Bohai Sea records.Discrepancies with historical records arise from methodological differences in species detection efficiency,the unique location of the ranch,and interannual variations in fish distribution;(3)Seasonal variations revealed maximum diversity in winter(60 species)and minimum in summer(30 species),with 12 species present year-round.Alpha diversity analysis showed higher diversity in autumn and summer than in winter and spring.Beta diversity indicated greater seasonal community differences than spatial variations within seasons.(4)The local database,despite having lower species coverage,demonstrated superior annotation accuracy compared with the NCBI database,thereby effectively eliminating non-target species interference and correcting misannotations for five species.The eDNA approach overcomes traditional limitations in reef areas(low catch efficiency and gear damage)while maintaining ecological integrity through non-invasiveness.This technique enables comprehensive monitoring of diverse ecological types and behavioral habits,providing robust data support for biodiversity conservation management in Laizhou Bay and effective technical evaluation for marine ranch restoration,thereby facilitating scientific management and sustainable development of marine ranching.
