Abstract

We investigated the sequence characteristics of the pmela gene in Japanese flounder(Paralichthys olivaceus)and its expression pattern in body color formation by cloning the CDS region of the pmela gene and analyzing its protein properties using bioinformatics.The expression profiles during larval pigment formation in different juvenile tissues and colored adult skin were analyzed via quantitative real-time PCR(qRT-PCR)and immunohistochemistry.The results showed that the CDS region of pmela was 2445 bp in length,encoded 814 amino acids,and contained a PKD domain and transmembrane region.Phylogenetic analysis revealed its closest evolutionary relationship with Pleuronectiformes species,sharing over 80%homology,and classified it as a stable hydrophobic protein.QRT-PCR analysis showed that during larval development,pmela mRNA expression levels differed synchronously with pigment deposition,indicating an initial upregulation followed by a later downregulation.Among different tissues,pmela was predominantly highly expressed in the eyes and skin.In adult skin,its expression levels in both gray and black skin were significantly higher than those in white skin(P<0.05).Immuno-histochemical analysis revealed that the Pmela protein was primarily enriched in the retinal pigment epithelium and the basal layer of the skin,with the highest distribution in black skin.The findings of this study confirm that the pmela gene participates in body color formation in Japanese flounder by regulating melanosome maturation.It is hypothesized that pmela may play a key role in the early stages of melanosome formation,laying a preliminary research foundation for obtaining golden-colored Japanese flounder through pmela gene editing.
