Abstract

To compare the distribution of Bacillus strains with cellulolytic potential from different regions and sources,and to identify highly efficient cellulose-degrading strains;Samples of animal feces,soil and humus,and biogas slurry and com-post were collected from Chengdu,Hongyuan,Kunming,and Xishuangbanna districts.After enrichment and isolation on CMC-Na medium,Bacillus strains were identified by 16S rDNA sequencing,and their cellulase activity was determined u-sing the DNS method;Among the 206 collected samples,a total of 1246 Bacillus isolates were identified as Lysinibacillus,B.subtilis,B.cereus,Brevibacillus,B.amyloliquefaciens,Paenibacillus,B.licheniformis,and B.altitudinis.The greatest species diversity was observed in soil and humus samples,while the highest abundance was from biogas slurry and compost,and the lowest diversity from fecal sources.Among soil-derived isolates,Chengdu yielded the highest number,Kunming the greatest species diversity,and Hongyuan the least in both aspects.At 45℃,both the isolation rate and species diversity of Bacillus were markedly higher.Of 193 strains tested for cellulase activity,171 were positive,with B.subtilis showing the highest mean enzyme activity(24.07 U·mL-1),while B.amyloliquefaciens and B.licheniformis showed the highest posi-tivity rate(100%).14 strains exhibited higher cellulase activity than the reference strain B.subtilis ACCC 11089,with strain B.subtilis FHYH58-6 reaching the highest activity(33.32 U·mL-1);The distribution of Bacillus strains with cellulo-lytic potential varies across regions and sources.Cellulase activity is strain-specific,with B.subtilis showing superior per-formance.Notably,strain B.subtilis FHYH58-6 exhibited the highest activity,highlighting its potential for further research and application.
