Abstract

Objective:To investigate the intervention mechanism of acupuncture on visual cortical plasticity in rats with monocular vision deprivation by examining the regulatory effects of"Tiao Qi Ming Mu"(Qi-regulating and eyesight-improving)on the metabotropic glutamate receptor 1(mGluR1)-mediated cyclic-adenosine monophosphate(cAMP)/protein kinase A(PKA)signaling cascade. Methods:Forty-eight Sprague-Dawley rats were randomly divided into a blank group,a model group,a covering group,and an acupuncture group.The deprivation amblyopia model was established by right eyelid suture in all groups except the blank group.In the covering group,the sutured right eye was cut open,and the left eyelid was sutured on the 7th day after modeling.In the acupuncture group,the"Tiao Qi Ming Mu"point prescription was administered starting on the 7th day after model establishment and continued for 14 d.The electrophysiological changes in binocular vision of rats were detected using pattern visual evoked potential(P-VEP);in visual cortex area 17,the mGluR1 protein expression was detected using immunofluorescence staining,the mGluR1 mRNA level was detected by real-time polymerase chain reaction,and the mGluR1,cAMP,and PKA protein levels were detected by Western blotting. Results:Compared to the blank group,the P-VEP's P100 wave latency period delayed,and the amplitude reduced in the vision-deprived eye of the model group,showing statistical significance(P<0.05);the protein and mRNA expression of mGluR1 and the protein expression of cAMP and PKA decreased in visual cortex area 17 in the model group,and the differences were significant(P<0.05).After 14 d of intervention,compared to the model group,the vision-deprived eye's P-VEP waveform improved with advanced P100 wave latency periods and increased amplitudes in the covering group and the acupuncture group,and the differences were significant(P<0.05);the fluorescence expression of mGluR1 protein and the protein and mRNA expression of mGluR1 increased significantly(P<0.05);the cAMP and PKA protein expression increased significantly(P<0.05)in the covering group and the acupuncture group. Conclusion:"Tiao Qi Ming Mu"point prescription can activate the cAMP/PKA signaling pathway mediated by mGluR1 in visual cortex area 17 and promote the synaptic plasticity changes of visual cortex neurons of monocular vision-deprived amblyopia model rats,thereby improving the optic nerve conduction efficiency and promoting the recovery of visual function.
